Abstract
Clonal selections of waratah were initiated into tissue culture.
The success of initiation and establishment of cultures was dependant on the maintenance of stockplants under protected cultivation for several months prior to explanting.
Adequate multiplication was achieved using a modified Murashige and Skoog (1962) medium.
Adventitious roots could be initiated on microshoots in vitro or in vivo. Acclimatisation of in vitro rooted shoots was not achieved.
Direct rooting of shoots of several genotypes was achieved using rooting powder (Seradix® N°2) and maintaining the shoots under high humidity in a fog regime (95–98%) until shoot growth recommenced.
