Abstract
A rapid, simple and improved extraction procedure for the detection and identification of viroids and plant viruses by dot-blot hybridization is described.
The entire tissue extraction can be carried out in a single tube and can be used to detect viroids or viruses in a number of plant tissues; i.e., leaves, stems, tubers, shoots, roots and seeds.
The extracts are blotted onto a nitro-cellulose membrane and incubated with 32p-labeled complementary DNA (cDNA) probes to detect and identify the infectious agent.
The sensitivity of detection is comparable to that achieved with phenol-extracted nucleic acids.
