EFFECT OF DIFFERENT FILMS USED FOR FILM CULTURE VESSELS ON PLANTLET DEVELOPMENT OF PHALAENOPSIS AND CYMBIDIUM
Therefore, we examined which film is more suitable for film culture vessel.
The plantlet development of Phalaenopsis grown on agar medium in CultuSAK™ (6" x 9", 5 chamber unit), in the Culture Bag of nearly the same size and shape, and in the test tube (25 mm x 150 mm) covered with aluminum foil was observed.
The plantlets grown in the Culture Bag were normal and larger when compared to those in CultuSAK™ and in the test tube.
To examine which film is more suitable for the box-shaped film vessel, sixteen shoots of Cymbidium and one-leaf of Phalaenopsis seedlings were cultured on agar medium in standard size Culture Pack (7.5 x 7.5 x 10.5 cm) made of 25 μm Neoflon PFA film and 31.8 μm high density polyethylene film, and in flask covered with aluminum foil.
The plantlets of both orchids grown in the Culture Pack made of Neoflon PFA film were larger and more vigorous compared to those grown in the other culture vessels.
This could be due to the higher light transmittance and better gas permeability of Neoflon film compared to the counterpart.
These results indicate that the Culture Bag and the Culture Pack made of Neoflon PFA film are recommended for use in plantlet development in micropropagation of orchids.
