ANALYSIS OF A TRANSPOSABLE ELEMENT CARRYING STREPTOMYCIN RESISTANCE IN ERWINIA AMYLOVORA

Abstract
Streptomycin resistance in Erwinia amylovora strain CA11 is located on a 34-kb plasmid (designated pEa34) and a 2.7-kb restriction fragment from pEa34 hybridizes with a portion of the streptomycin resistance gene (probe SMP3) from strain Psp36 of Pseudomonas syringae pv. papulans. The nucleotide sequences associated with a streptomycin resistance in E. amylovora strain CA11 and P. s. papulans strain Psp36 were identical, except for 3 bp, to the strA and strB genes found on the nonconjugative plasmid RSF1010. The resistance genes in E. amylovora were carried on a Class II, Tn3-type transposable element, designated Tn5393, that moved from pEa34 to different sites in plasmid pGEM3Zf(+). Also, transposon Tn5393 was common to most gram-negative bacteria found in apple orchards with strA and strB genes.
Transposition and conjugation are the likely mechanisms for the widespread distribution of strA and strB among gram-negative bacteria.
