ISOLATION, IDENTIFICATION AND DETECTION OF UNDESCRIBED RNA SWEETPOTATO VIRUSES

Abstract
Sweetpotato cultivar decline has viruses among the causal agents of this complex disorder.
The isolation and characterization of these viruses are difficult due to the presence of sweetpotato feathery mottle virus (SPFMV) in most plants.
Recently we initiated a study to determine the presence of undescribed viruses infecting sweetpotatoes in North Carolina.
A ds-RNA-RT-PCR-Cloning and sequencing based novel approach is being used for molecular identification and characterization of unknown viruses.
Initially several symptomatic sweetpotato plants were tested by serology.
Most of them were SPFMV positive.
From the few negative samples a plant from the cv George Wooten Beauregard showing strong symptoms and a unique ds-RNA profile was selected for cloning and sequencing.
Sixteen clones were sequenced and analyzed.
Phylogenetic analysis showed the highest homology with SPFMV; two with SPV-G and four are listed, as unknown Potyvirus From the last group 2 clones are closer to SPMMV. Our results show that this new approach will be useful in detecting and characterizing undescribed sweetpotato viruses.
