IN VITRO PROLIFERATION OF OLIVE (‘DOLCE AGOGIA’ AND ‘MORAIOLO’): EFFECT OF DIFFERENT CYTOKININS

Abstract
In tissue culture the growth regulators represent the main factors to promote the proliferation.
In order to individuate an effective hormonal combination and to contain the high costs of the media components, the effect of two cytokinins was investigated on the proliferation of vitro-derived explants of two olive cultivars (Dolce Agogia and Moraiolo). Uninodal explants were taken from proliferated shoots and were vertically cultured on 100 ml of medium containing 2 and 4 mg L-1 of zeatin and kinetin, alone or in combination.
After three subsequent subcultures, the influence of cytokinins was evaluated.
The data analysis indicated that the proliferation activity was greatly influenced by the hormonal component and the genotype.
In fact, Moraiolo showed the most satisfactory levels of multiplication rate under the effect of 2 and 4 mg L-1 of zeatin (8.2 and 8.8 respectively). Interesting results were also obtained in Dolce Agogia because significant sinergistic effects on the proliferation activity were recorded in the presence of zeatin and kinetin in combination at the lowest concentration (8, 4). These results contributed to improve the effectiveness of the in vitro proliferation protocol in olive.
