Abstract
Genomic library construction for the two species of dragon fruits, Hylocereus undatus and Hylocereus polyrhizus, using bacteriophage Lambda Fix II with 20 kb inserts was reported.
About 80-100 plaques were observed for each plate, indicating that bacteriophage has lysed the host cells, XL1-Blue MRA (P2). Best methods for genomic library construction were discussed in details.
These include total DNA digestion using restriction enzyme, BamHI, phenol-chloroform purification of the concentrated insert DNA, ligation, packaging, and titering the packaging reaction.
