MICROPROPAGATION OF CHESTNUT (CASTANEA SATIVA MILL.) IN SEMI-SOLID CULTURE

Abstract
In this study, micropropagation of three different chestnut genotypes were compared in the Gresshoff and Doy (GD)+Murashige and Skoog (MS) (½ NH4NO3 and KNO3) and MS (½ NH4NO3 and KNO3) semi-solid media.
Genotypes used in this study were selected among chestnut stands from the Nazilli district of Aydın province.
The genotype numbers are N-3-4, N-23-1 and N-7-3. During the experiments, chestnut buds cultured in January in GD medium were transferred to MS (½ NH4NO3 and KNO3) medium after six weeks in March.
The development of buds in this culture was observed for a six-week time period.
In March, another set of buds were directly planted onto MS (½ NH4NO3 and KNO3) medium and they also were observed for six weeks.
From our results, the shoot formation level reached 41.48% in MS (½ NH4NO3 and KNO3) medium and 25.51% in GD+MS (½ NH4NO3 and KNO3) medium.
The highest value of development was obtained with offshoots from the N-3-4 (37.21%) genotype in MS (½ NH4NO3 and KNO3) medium.
The number of leaves of the chestnut genotypesaveraged 6.47 leaves in MS (½ NH4NO3 and KNO3) medium and 5.16 leaves in GD+MS (½ NH4NO3 and KNO3) medium.
