Abstract
The genetic variability of Xanthomonas campestris pv. campestris isolates mainly from the Czech Republic was investigated.
This plant pathogen contains a type III secretion system (TTSS) that is necessary in order to attack the host.
TTSS is important in pathogenesis because it transfers effector proteins in order to lower the host’s defence.
The hrpF gene is therefore an essential gene in the pathway of the TTSS. Zur gene is a regulator with high-affinity uptake of zinc in many bacteria like Xanthomonas campestris pv. campestris. The Zur gene regulator family controls homeostasis of zinc, EPS (extracellular polysaccharide) production, and virulence in X. campestris pv. campestris. In this study, primers were designed, and using them the Zur gene was partially amplified.
In this study both obtained amplicons of hrpF and Zur genes were sequenced, and based on sequencing analysis the phylogenetic relationships were determined.
