Simple and efficient method for the extraction of genomic DNA from litchi

Abstract
The isolation of good quality DNA is the prerequisite for molecular research in any plant.
However, the presence of high polyphenols, tannins and polysaccharides contents in leaf tissue of litchi leaf makes the genomic DNA extraction more difficult.
In this study an efficient and sustainable method of genomic DNA extraction procedure is developed.
This protocol is an improvement over the existing CTAB methods.
Fully expanded, fresh, tender and purplish/reddish brown leaf was used to extract quality DNA. The addition of polyvinylpyrrolidone (PVP) and mercapto-ethanol was effective in the elimination of secondary metabolites, particularly polyphenolic compounds.
The incorporation of high salt washing step was also efficient to remove polysaccharides.
Therefore, results from electrophoresis, ratios of A260nm and A280nm and marker studies suggested that this modified method is more efficient to extract high-quality DNA compared to other reported conventional methods.
