Celeriac protoplast regenerated plants phenotypic and genotypic characterization

Abstract
Protoplast regenerated plants have an increased risk of accumulation of somaclonal variability, making the assessment of the regenerated material necessary.
We analyzed the flower morphology and pollen viability and we determined the chromosome number of a set of celeriac plants comprising both protoplasts and cell suspension regenerants.
We observed consistent abnormal anther formation and lack of pollen for one of the protoplast lines (line 3.1) but not for the other protoplast line (line 3.2) or cell suspension derived plants.
Alexander staining of the pollen exposed the preponderant presence of mixed anthers with both fertile and sterile pollen in plants regenerated from cell suspension and protoplast line 3.2. Out of 17 regenerants, one protoplast regenerant was a mixoploid of diploid and aneuploid cells and 2 protoplast regenerants contained diploid and tetraploid cells indicating a low frequency of genetic instability in celeriac plants regenerated from protoplasts.
