Expression of genes associated with ‘Benny’ Valencia rind pitting

Abstract
Rind pitting is one of the most important disorders of citrus fruit that affect the quality and decrease the market value of citrus due to deterioration of the rind.
The expression levels of gene in the peel during the development of rind pitting of ‘Benny’ Valencia fruit have been investigated. ‘Benny’ Valencia fruit were harvested from Tzaneen, Groblersdal and Musina in South Africa.
Afterwards, fruit were separately subjected to the following treatments as: no wax with dehydration, wax with dehydration and wax without dehydration.
Dehydrated treatments were applied for 3 days at relative humidity of ±45%, thereafter; fruit were stored at -0.6 and 4.5°C for 28 days plus 7 days of shelf-life.
After RPI analysis, the flavedo was removed, store at -21°C and was used to screen differentially expressed genes in citrus rind pitting by quantitative RT-PCR method.
The pitting rind and no-pitting rind were selected as the tester and driver, respectively.
Three homologic genes: CsCP gene; CsNAC-domain protein gene; CsCP-F gene; were chosen to examine the relationship between their expression and citrus rind pitting.
The expression of those genes were all higher in the pitting rind fruit harvested from Tzaneen and low in fruit with low pitting.
Groblersdal and Musina fruit had low expression of genes and low rind pitting was also observed.
The results suggest that gene expression may be linked with rind pitting and could serve as targets for future investigation.
