Abstract
Due to a high level of virus disease in the major raspberry cultivars grown in Czechoslovakia, a virus elimination program involving indexing and meristem culture has been developed.
Indexing is done by sap inoculation to cucumber and by ELISA. A special combined method of meristem culturing involving thermotherapy and repeated meristem culturing eliminated virus from more red raspberry cultivars than did one meristem culturing.
Seven temperature regimes were tested.
Those in which the explant is grown at ambient temperature for at least 14 days and then exposed to a prolonged period of heat therapy at 37 C were most successful in eliminating viruses and allowing survival and development of explants.
