IDENTIFICATION OF A RAPD MARKER TIGHTLY LINKED TO A SELF-INCOMPATIBILITY GENE IN NON-HEADING CHINESE CABBAGE

Abstract
Bulk segregant analysis (BSA) and random amplified polymorphic DNA (RAPD) were employed to identify molecular markers linked to a self-incompatibility gene in self-incompatible and self-compatible near-isogenic lines of non-heading Chinese cabbage (B. campestris L. ssp. chinensis Makino var. communis Tsen et Lee). Primer S1107 produced a 900 base pair (bp) reproducible fragment only in the self-incompatible line.
Using the F2 mapping population from a cross of SC104-1 x SI13-2, S1107-900 was shown to be closely linked to the self-incompatibility gene at a distance of 7.54±0.11cM. This work provides a basis for molecular marker-assisted selection for self-incompatibility that should lead to localization and cloning of the controlling gene.
