STABILITY OF SHORT SEQUENCE REPEATS AND SUITABILITY OF SSR, AFLP AND RAPD FOR THE CHARACTERIZATION OF ERWINIA AMYLOVORA STRAINS

Abstract
In this study we demonstrated that the short sequence repeat (SSR) marker of the pEa29 plasmid of Erwinia amylovora fulfils the requirement for a genetic marker.
Therefore, we determined the number of SSR units of several E. amylovora isolates for further characterization and for epidemiological purposes.
Using GAATTACA as the repeat unit, we found SSR numbers from 3 to 15 in isolates from Hungary, Germany, and Austria.
This is one unit more than described previously.
For strains in culture collections, the differentiation into PFGE-type (Pt1 to Pt6) and SSR-type (SSR-3 to SSR-15) represents a valuable tool for strain classification yielding 78 distinct E. amylovora types.
Random amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP) primers were tested for their suitability for differentiating E. amylovora strains and to support the data obtained by SSR analysis.
