IN VITRO CULTURE OF SEVERAL ORNAMENTAL AND MEDICINAL SALVIA SPECIES

Abstract
In the context of an international project on the valorisation of the genus Salvia both for ornamental and medicinal purposes, several Salvia species were introduced in vitro for propagation studies and with the aim to produce biomass for extraction of secondary metabolites.
Only a few reports on the in vitro propagation of the genus Salvia were found in literature, and not one in the species considered in this work.
In the present paper, data about sterilization are reported together with results on micropropagation efficiency.
In order to obtain aseptic material, axillary and apical buds of adult plants of 19 Salvia species grown in the greenhouse were used as explants; for most of them it was necessary to use two pre-treatments (ethanol 70%, 30 sec and 40 °C water with few drops of liquid soap) before use of sodium hypochlorite.
It was possible to reach sterilization percentages of up to 50 %, with high viability among the 19 species considered.
A screening of the multiplication rate was made with 8 species in the presence of BA (0.3 mgL-1 ) and in particular the species S. elegans, S. sinaloensis, S. cinnabarina and S. jamensis “la Siesta” were used for an experiment on multiplication efficiency and quality of the produced plants related also to the acclimatization phase.
A high variability was defined among the species; in vitro multiplication of Salvia is better mediated by a low level of BA. Kinetin did not sufficiently increase the multiplication rate.
The plant quality (height and weight) can be modulated in order to obtain plants with good acclimatization performances.
