FLOWER COLOR AND FLORESCENCE MUTANTS OBTAINED USING ELECTRON BEAM IRRADIATION OF CHRYSANTHEMUM BUDS

Abstract
Enrichment of flower color and advance or delay flowering are the main objectives in chrysanthemum breeding.
In this study, electron beam irradiation combined with in vitro culture was used to develop a new mutation breeding method in chrysanthemum.
Buds of orange flower ground-cover chrysanthemum ‘Cheng Yun’ were treated with electron beam irradiation at 20, 40, 60 and 80 Gy.
The irradiated buds were dissected and their florets were inoculated onto Murashige & Skoog (MS) media supplemented with sucrose and for callus induction.
The well-grown calli were subcultured onto MS media but with different combinations of NAA and BAP for shoot regeneration.
Results showed that the optimal medium for callus induction was supplemented with 1.0 mg/L NAA and 2.0 mg/L BAP while the optimum medium for shoot regeneration contained 0.2 mg/L NAA and 1.0 mg/L BAP. The regenerated shoots rooted well in half MS basal medium.
The rooted shoots were transplanted into pots containing sterile perlite and vermiculite mixture (1:1). It was observed that flower color and day to flowering of some mutants were altered by the electron beam treatment.
The dose of 40-60 Gy was optimal for inducing mutations.
The altered flower colors ranged from yellow to red-purple and the day to flowering was 25 days earlier or 15~20 days later than that of the control.
The mutants were maintained vegetatively and produced true-to-type flowers.
