SIMPLE SEQUENCE REPEAT ASSESSMENT OF GENETIC DIVERSITY AMONG WILD POPULATIONS OF CHINESE CHESTNUT

Abstract
We surveyed the variation in microsatellites in sixty-nine accessions of Chinese chestnut (Castanea mollissima) collected from various wild populations in China.
We found ten simple sequence repeat loci that showed eighty-four variations in the tested plants.
The number of alleles per locus ranged from four-to-fourteen (average, 8.4). The effective Ne of alleles was 4.998. Average values for expected heterozygosity and polymorphism informative content were up to 0.777 and 0.739, respectively.
The highest genetic diversity was found in the Shanxi population.
The low value for genetic differentiation (Gst=0.141) showed that most of the genetic variation was within populations.
A UPGMA phenogram showed that the populations from Yunnan, Chongqing and Anhui provinces could be classified into two groups; principal coordinate analysis showed the same result.
The two methods showed that there was not a significant correlation between genetic relationships and geographical distance among the wild populations tested.
