ESTIMATING GENETIC DIVERSITY IN NEW GUINEA IMPATIENS USING AFLP MARKERS

Abstract
The amount of genetic diversity present in 41 cultivars of New Guinea impatiens (Impatiens × hybrida) was evaluated using amplified fragment length polymorphism (AFLP) technology. A total of 48 EcoR I and Mse I primer combinations were used. The AFLP analysis generated between 93 and 153 scorable bands per primer combination. A dendrogram was generated from the AFLP information that revealed six groupings among these 41 cultivars. Two different primer labeling techniques, radioactive and fluorescence were also evaluated. AFLP profiles of three cultivars revealed that levels of polymorphism using fluorescent-labeled primers were similar to those observed using radioactive-labeled primers. The implications of these findings on genetic relationships and genetic diversity in the New Guinea impatiens genome will be discussed.
