MOLECULAR DIVERSITY IN POPULATIONS OF MALUS SIEVERSII (LEDED.) ROEM.

35 wild Malus sieversii offsprings from 10 collecting areas in Kazakhstan were selected and germinated for evaluation at the former fruit genebank in Dresden-Pillnitz, Germany (in total 1063 seedlings, material collected by USDA-ARS Plant Genetic Resources Unit). For molecular diversity studies, a fluorescence-based variant of the AFLP technique was adapted for Malus on automatic fragment analyzers.
Six out of 208 primer combinations were selected and applied to all individuals, aiming at a detailed description of the molecular diversity within the material, followed by the development of a core collection for further conservation.
Preliminary results on diversity in 67 Malus sieversii individuals plus six additional Malus species show a partial clustering of the individuals from the same offspring and from the same area of origin, an unstructured grouping of other individuals, as well as an intermediate position of some M. sieversii samples from a specific collection area between all other M. sieversii individuals and the additional Malus species.
