ANALYSIS OF IN VITRO PERFORMANCES AND GENETIC VARIABILITY OF SEVERAL CAMPANULA SABATIA POPULATIONS IN ITALY

Savona’s bell (Campanula sabatia De Not.) is endemic to NW Italy (Liguria) and owing to the heavy human pressure it was included in the European Red list of endangered species.
Nine populations have been recognised in the Western Italian Riviera and seeds were collected during the summer period; the evaluation of germination percentages and mean germination time (M.G.T.) was performed in vitro. 20 seedlings of each population have been cloned in a medium composed by the basal Murashige and Skoog (1962) salt and sugar composition plus 6-BA 1.33 µM. Data on multiplication and rooting aptitude, the height of the clusters and the quality of the plants were scored for each clone and valuated as frequencies.
A large variability of the evaluated parameters was detected inter and intra each population.
An AFLP-based approach was used to assess the molecular genetic variation.
The DNA was successfully extracted from 10 different individuals for each population.
The preliminary screening of AFLP primers was performed on a few genotypes: 6 primer combinations, each able to produce more than 25 polymorphic bands, were selected.
