CROSS AMPLIFICATION OF ONION DERIVED MICROSATELLITES AND MINING OF GARLIC EST DATABASE FOR ASSESSMENT OF GENETIC DIVERSITY IN GARLIC

Abstract
Garlic (Allium sativum L.) is one of the most important vegetable alliums consumed worldwide for its food and medicinal uses.
Assessment of genetic diversity based on molecular markers is important since it is unaffected by the environmental conditions and reveals the true genetic nature of the germplasm.
Ninety three garlic accessions collected from different parts of the country and abroad were used in the present study.
Sixty genomic and EST based onion microsatellites, 8 newly reported garlic microsatellites and 31 newly mined microsatellites were used for amplification.
Only 8% of onion microsatellites were cross transferable whereas 88% of the garlic microsatellites were amplifiable.
Out of 31 newly mined SSRs, 35% were amplifiable in garlic germplasm.
Here we characterized 18 polymorphic SSR markers for assessment of genetic diversity in garlic germplasm.
Considerable genetic diversity was observed in the germplasm and it was observed that cluster formation was independent of the geographical location of the material.
We found that clustering was mostly on the basis of vernalisation requirement for bulb formation in garlic.
Till now only 8 garlic SSRs have been reported and we are reporting 18 SSRs useful for future studies on diversity assessment, core set construction and garlic germplasm identification.
