STEM-INDUCED CALLUS INDUCTION AND IN VITRO CULTURE OF FIVE ORNAMENTAL HERBACEOUS PEONY CULTIVARS

Abstract
The induction of callus plays an extremely important role in the protection of plant resources and genetic transformation.
This study aimed to explore the effects of medium, plant growth regulators and culture conditions on callus induction of five herbaceous peony (Paeonia lactiflora Pall.) cultivars: Da Fu Gui, Sorbet, Coral Sunset, Duchesse de Nemours and Sarah Bernhardt. These results have wide-spread significance for peony micropropagation, quality improvement, germplasm preservation and production of important compounds.
Transverse sections of young stems of all five cultivars, which served as the explant source, were collected in April 2011 from 3- to 5-year-old plants.
The experimental results show that half-strength Murashige and Skoog (MS) was the best basal callus induction medium for Coral Sunset, Duchess de Nemours and Sarah Bernhard. Optimum callus induction medium was 0.5 mg/L thidiazuron (TDZ) + 0.5 mg/L α-naphthaleneacetic acid (NAA) for Da Fu Gui, Sorbet and Sarah Bernhard and that for Coral Sunset and Duchess de Nemours was 0.5 mg/L TDZ + 0.5 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D). The most suitable medium for callus proliferation was 0.5 mg/L 2,4-D + 0.5 mg/L TDZ for Du Fu Gui, 1.0 mg/L 2,4-D + 0.5 mg/L TDZ for Sorbet, 0.2 mg/L TDZ + 0.2 mg/L NAA for Coral Sunset, 1.0 mg/L 2,4-D + 0.5 mg/L TDZ + 0.5 mg/L NAA for Duchess de Nemours and 1.0 mg/L 2,4-D + 0.5 mg/L TDZ + 0.2 mg/L NAA for Sarah Bernhard. The proliferation ratio of Duchess de Nemours was highest followed by Da Fu Gui, Sorbet, Sarah Bernhard and Coral Sunset. The callus of all five herbaceous peony cultivars was unable to differentiate into adventitious buds.
