TRANCRIPTOMIC STUDY OF FLOWER PETAL SENESCENCE IN GARDENIA JASMINOIDES ELLIS USING ILLUMINA HI-SEQ TECHNOLOGY

Abstract
Gardenia is one of the most important ornamental plants in Greece and has a great economic value for the country’s floriculture.
Its pure white flowers are highly fragrant which, however, have short life span.
In the present work we used large scale deep sequencing technology in order to study flower petal senescence at four floral stages: A) closed flower bud ready to open, B) flower that has just opened, C) fully opened flower and D) flower with signs of petal senescence.
Buds/flowers from the above stages were collected and total RNA was isolated from the petals and sequenced with the platform Illumina HiSeqTM 2000. Obtained data were analyzed and the resulting unigenes were annotated according to their function.
Transcripts were found which expression significantly differed at sequential floral stage comparisons and emphasis was given on the transcription factors and how their gene expression changed during the progress of the phenomenon.
According to the results, 107 unigenes that belong to transcription factors were differentially expressed during stage comparison A-B. For the stage comparison B-C, 58 unigenes belonging to transcription factors were differentially expressed, while for the stage comparison C-D the number increased to 81 unigenes.
These results show the complexity of the phenomenon of gardenia flower petal senescence which needs further research focused on specific transcription factors.
