Effects of stress from Aphis gossypii Glover on malondialdehyde content and protective enzymes activities in cucumber

Abstract
Cucumber is one of the most important vegetable crops in the world.
The aphid, Aphis gossypii Glover, is one of the main pests, which often causes significant damage on cucumber and reduction in yield.
Malondialdehyde (MDA) content and activity of peroxidase (POD), superoxide dismutase (SOD), catalase (CAT), phenylalanine ammonialyase (PAL), polyphenol oxidase (PPO) change in plants as a response to stress.
In order to analyze the defensive role of MDA and protective enzymes against aphid in cucumber, cucumber lines EP6392 (resistant) and JY30 (susceptible) were measured for their MDA content and protective enzymes activities.
Five apterous adult aphids were transferred to the back of the first true leaf of each cucumber seedling plant as treatment and the seedling plant without aphids was used as control.
The results indicated that MDA content and POD, PAL activities in seedlings infected by aphid were always higher than in control plants of both lines.
MDA content increased initially after aphid infestation and then declined in both two lines.
However, the MDA content of JY30 was higher than in EP6392. POD activity of the resistant line was increased more rapidly than that in the susceptible line.
PAL activities also increased after aphid infestation and then declined, and that in the resistant line was higher than that in the susceptible line after aphid infection.
Therefore, these results indicated that the MDA content, POD and PPO activities are likely to be involved in aphid-resistance in cucumber.
