Some special features of the conservation of valuable, essential oil rose cultivars: in vitro deposition and cryopreservation

Abstract
Plant genetic resources conservation of species is important for the production of essential oil, aromatic compounds, medicinal products and decorative properties.
The aim of our study was to optimize the conditions for long-term deposition and cryopreservation of valuable cultivars of different essential oil cultivars of rose grown in the south of Russia.
The regenerants of essential oil rose were cultured on MS medium with 0.1 mg L-1 NAA, 0.5 mg L-1 GA3, 0.5-1.5 mg L-1 BAP, 30 g L-1 sucrose and 8 g L-1 agar at 25±1°C under a 16-h photoperiod with a light intensity 37.5 μmol m-2 s-1. Explants for deposition were placed on ½ and ¼ MS media with 0.2 and 0.4 g L-1 CCC and 60 g L-1 sucrose.
Preservation was performed at low positive temperature (from 4 to 14°C) under 16-h photoperiods supplied by cool-white fluorescent lamps giving 1.25 μmol m-2 s-1. Microshoots for meristem isolation were hardened off for 2 weeks before cryopreservation.
The encapsulation-dehydration method was used for cryopreservation; using 3% alginate solution in MS medium with 0.75 M sucrose, polymerized in calcium chloride solution, dehydrated in MS medium with 0.75 M sucrose and dried under laminar flow hood to 20% humidity). Regeneration viability and frequency were assessed on the 30th day after thawing during culture on the regeneration medium.
Multiplication index in the long-term collection in vitro was 1:3-1:6, the maximum in the cultivars ‘Raduga’ and ‘Festivalnaya’, respectively.
Microshoot height and leaf amount were less under the deposition, leaf blade morphology differed with those under the standard in vitro culture.
After the cryopreservation, the viability of the studied samples varied from 42 to 81%. The efficiency of regeneration did not differ significantly from the control.
In some cultivars we observed an increase in leaf number, adventitious microshoots appearance and active rhizogenesis.
