Abstract
Hibiscus moscheutos is a native herbaceous perennial plant.
Because it is an ornamental plant, variations in aesthetic traits are valuable.
Mutagenesis is a method to increase variation in hibiscus plants, and gamma irradiation is a common physical mutagen that causes changes to the genome.
The effects of various levels of gamma irradiation on hibiscus plants were analyzed.
Seeds from cultivar ‘Luna Red’ were used for treatment, as it is an inbred line.
The study was performed using a randomized complete block design with three replications of 15 seeds treatment‑1. Radiation treatments consisted of 0, 50, 100, 200, 400, 800, and 1600 Gy (gray). The germination percent was measured at 12 days after treatment (DAT), and the survival percent and height of seedlings were measured at 50 DAT. Data was analyzed in R Studio using a linear mixed-effects model for germination and survival, and an ANOVA and Tukey’s HSD for height.
Radiation level was found to cause a significant decrease in germination, seedling survival, and seedling height.
Plants treated with 1600 Gy had a germination percent that was half that of other treatments.
There was ~50% greater survival in the plants treated with 200 Gy than those treated with 800 and 1600 Gy, and ~35% greater survival in the plants treated with 200 Gy than those treated with 400 Gy.
Plants treated with 0, 50, and 100 Gy had ~100% greater survival than those treated with 800 and 1600 Gy.
Plants treated with 0 and 50 Gy were ~33% taller than those treated with 100 Gy, ~85% taller than those treated with 200 and 400 Gy.
The results of this study will enable the determination of optimal treatment levels for H. moscheutos, a species of cold-hardy hibiscus, using gamma irradiation.
