Abstract
As one of the key ancestors and also a widely used stock of modern rose, the eastern Asia originated Rosa multiflora is a very important germplasm resource for rose breeding and in rose industry.
Four varieties have been identified with rich morphological diversity within the species.
The population genetics of R. multiflora complex were studied in order to assess the genetic variation and to explore the possible evolutionary mechanism behind the differentiation of the complex by using three highly divergent chloroplast DNA (cpDNA) intergenic spacers.
A total of 31 cpDNA haplotypes were detected in 565 individual plants from 33 populations collected from the natural distribution in China and Japan, and the most dominant haplotype exists in 24 populations.
The complex showed a rather high genetic diversity (HT=0.851, Hd=0.750) and low genetic differentiation (GST=0.304). All the populations could be divided into five phylogeographical groups.
Most (67.23%) of the genetic variation was from among the groups, indicating a significant geographic barrier among the groups and a common gene flow among populations within groups.
The morphology-based classification of four intraspecific varieties was not supported by the phylogeny of cpDNA haplotypes. Rosa multiflora evolved as a species ca. 3.51-2.52 Mya, and first diverged ca. 3.02-2.28 Mya.
The ancient lineages mainly existed in the mountainous areas in southwestern and southern China during the Quaternary Climate Oscillations.
Populations Lushan, Ningqiang, Wushan, Changde, Tongren, Zhaotong, and Baise of China could be evaluated as the core germplasm banks of R. multiflora complex.
