Abstract
Eight monoclonal antibodies (MCAs) to Erwinia amylovora have been produced using two antigens: exopolysaccharides (EPS) of strain 1430 and whole cells of the acapsular avirulent mutant PMV 6089 of 1430. These MCAs have been tested in ELISA-DASI against 48 E. amylovora strains from different origins, 45 strains of other plant pathogenic bacteria and 258 bacterial strains from epiphytic microflora of rosaceous plants.
A different pattern of reaction was observed when using different MCAs.
Three MCAs reacted against all the E. amylovora strains assayed.
The majority of the MCAs were E. amylovora specific.
Two strains from apple epiphytic microflora reacted in ELISA-DASI with three MCAs, but no reaction was observed after boiling for 10 minutes.

Several serological techniques, useful for detection or identification of E. amylovora in infected plants or pure cultures, were assayed using selected MCAs against EPS and against Ea PMV 6089 strain.
Some MCAs reacted with high affinity in immunofluorescence, ELISA indirect, ELISA-DASI, immunoelectrotransfer (Western blot) and immunoprinting.
The ability of these MCAs to react in different serological techniques alone or in a mixture allows a specific and sensitive detection of E. amylovora.
