MOLECULAR TYPING OF ROOTSTOCK HYBRIDS (CASTANEA SATIVA X CASTANEA CRENATA) AND PORTUGUESE CASTANEA SATIVA CULTIVARS BASED ON RAPD MARKERS

Abstract
Chestnut is an important culture in Portugal, but the ‘ink disease’ caused by Phytophthora cinnamomi (Petri) and Phytophthora cambivora (Rands) has severely affected most regions in the north and centre of the country.
A collection of hybrid rootstock selections (Castanea sativa x Castanea crenata) is maintained at E.N.F.V.N. and a database of the fingerprinting of these rootstocks seemed a first step in the identification and maintenance of the collection with the minimal number of replications.
Thirty-nine accessions of Castanea: four C. sativa, one C. crenata and thirty-four hybrids of C. sativa x C. crenata were selected from the collection.
From a screening of forty-three 10-mer random primers, twelve primers generated 164 polymorphic bands.
Amplified products were analysed by comparing the genotypes based on the percentage of common fragments and a similarity matrix using the NTsys-pc program for cluster analysis.
There was a clear separation between C. crenata and both C. sativa and the hybrids (42.4% similarity). The Portuguese cultivars of C. sativa were separated from the hybrids forming a subcluster.
Within the hybrids, three subclusters were separated and ten pairs of hybrid genotypes showed similar or very similar patterns.
Besides typing, the RAPD results are a first approach of the phenetic proximity of the hybrids between themselves and in relation to the progenitor species.
