Abstract
Carnation etched ring virus (CERV) was partially purified from mechanically inoculated Saponaria vaccaria Pink Beauty leaves with butanol clarification alone or in combination with urea and Triton X-100. Infectivity was associated with a major UV absorbing band about 12 to 17 ml below the meniscus and in several fractions adjacent to the band in a 10 to 40% sucrose density gradient.
CERV nucleic acid (NA) consists predominantly of linear molecules that range in length from 0.12 μm to more than 35 μm with most of the strands less than 1 μm.
A few molecules were circular without crossovers with a molecular weight of about 4.5 X 106 daltons.
Cauliflower mosaic virus (CauMV) NA occurs predominantly as circular molecules with a unit length of 2.1–2.3 μm, similar to CERV. Some CauMV molecules are linear with the same unit length as the circular forms.
CERV-NA was resolved in some experiments in composite polyacrylamide-agarose gels into 3 fluorescent bands after ethidium bromide staining.
The resolution of the bands and staining intensity of the zones varied among experiments and only a rapidly migrating band was consistently observed.
CauMV-NA separated into at least 3 distinct electrophoretic components in composite polyacrylamide-agarose gels.
