Abstract
A protocol has been developed for in vitro plant regeneration from seed cotyledon of Camellia japonica L. Adventitious buds were efficiently formed without callusing on the proximal region of mature seed cotyledons cultured on Murashige and Skoog (MS) medium supplemented with 0.1 mg L-1 2,4-D and 1 mg L-1 BA. A prolifering shoot culture was established by repeatedly subculturing the original cotyledon explant on fresh medium after each harvest of the newly formed shoots.
Nearly 38% of the shoots formed roots after being transferred to MS medium without plant growth regulators.
These results suggest that cotyledon of mature seeds can be used as a source of explants for the experiments of clonal proliferation and genetic transformation in Camellia species.
