STUDY ON CHANGES IN PROTEIN PATTERNS AND PHENOLIC CONTENTS DURING SOMATIC EMBRYOGENESIS IN CROCUS SATIVUS L.

Abstract
Crocus sativus L. is an herbaceous plant belonging to the Iridaceae family.
Somatic embryogenesis was induced from shoot meristem culture on Linsmaier and Skoog medium containing 4 mg/L kinetin and 1 mg/L 2,4-D. To initiate morphogenesis, embyogenic calli were transferred onto 1/2 Murashige and Skoog medium containing 1 mg/L ABA. Plantlet regeneration was achieved by transferring germinated embryos to 1/2 MS medium containing 0.1 mg/LNAA and 1 mg/L BA. In order to understand the embryo maturation process, changes in protein patterns during somatic embryogenesis were studied by SDS-PAGE method.
Results showed that the activity of protein synthesis increased during the first four days.
Also the presence of 20, 31 and 50 KD proteins in the heart-shaped embryos and more developed ones were distinguished.
The synthesis and deposition of these storage and LEA proteins during somatic and zygotic embryogenesis are usually regulated through ABA- and water-stress-induced gene expression.
Also polyphenol oxidase and peroxidase activities were studied by PAGE method.
Results showed the increases in activities of these enzymes during embryogenesis.
Monophenolic acids also increased in the early phases of somatic embryogenesis and then decreased during embryo development.
These compounds might influence through lowering the endogenous IAA.
