IN VITRO PROPAGATION OF HYBRID CANDIDATE ROOTSTOCKS FOR ORNAMENTAL CITRUS

Abstract
Some hybrids obtained in 2005 and 2006 crossing chinotto small leaf (Citrus myrtifolia Raf.) with trifoliate orange [Poncirus trifoliate (L.) Raf.], Flying Dragon trifoliate orange [P. trifoliate var. monstrosa (T. Ito) Swing.] and Troyer citrange (P. trifoliata × C. sinensis) were developped with the aim to test them as candidate rootstocks able to limit the canopy growth for ornamental use.
The long juvenile period (at least 5 years to start blooming) makes conventional breeding in citrus unusually slow.
The aim of this work is to establish a procedure in vitro culture to shorten this time by propagation system.
Some budsticks of six greenhouse-grown hybrids were used as the source of 20-30 nodal segments 1.0-1.5 cm in length and cultured in MS containing 3 mg/L
6-benzylaminopurine.
Three weeks later the shoots were transferred to a medium multiplication containing 0.5 mg/L of 6-benzylaminopurine and 0.25 mg/L of indole-3-butyric acid and, 60 days after, in an elongation medium including 1.0 mg/L gibberellic acid.
Root formation was subsequently obtained in a mix of peat and expanded perlite.
At present several saplings of each rootstock are cultivated under greenhouse to be grafted within one year.
