Abstract
Yuzu (Citrus junos) is the most popular commercial acid citrus grown in Japan; however, the botanical origin of Yuzu as hybrid or non-hybrid Citrus species is confusing and controversial.
In order to clarify the hybrid origin of Yuzu and its relationship with other acid citrus cultivated in Japan, two experiments were carried out in this study, in the first; PCR-RFLP analyses of three regions for each of chloroplast DNA (cpDNA; rbcL-ORF106, trnD-trnT, trnH-trnK) and mitochondrial DNA (mtDNA; nad7/exon2-exon3, nad7/exon3-exon4, 18s-5s) were performed in 26 cultivars of acid citrus, three true Citrus species, i.e., mandarin, pummelo and citron; and one papeda, ichang papeda (C. ichangensis). In the second experiment those two unique cpDNA restriction patterns of Yuzu found in one region of chloroplast DNA (trnH-trnK) were assessed in 22 mandarins to find their maternal relationship.
Based on the results of this study only four cultivars of acid citrus, Matsuda Sudachi, Zuishoyu, Hanayu and Yuko showed both cpDNA and mtDNA restriction patterns as same as Yuzu.
Also, the two unique cpDNA restriction patterns of Yuzu were not observed in the mandarin accessions, therefore the hybrid origin of Yuzu was not confirmed in this study.
The data revealed that Yuzu acted as female progenitor in the origin of four acid citrus grown in Japan.
