MOLECULAR CHARACTERISATION OF ‘CANDIDATUS PHYTOPLASMA ASTERIS’ AND ‘CANDIDATUS PHYTOPLASMA MALI’ ASSOCIATED WITH LILY LEAF SCORCH AND FLOWER BUD ABSCISSION

Abstract
The occurrence of upper leaf necrosis and malformation as well as flower bud abscission were observed in naturally infected oriental lily hybrids Simplon and Siberia cultivated in commercial greenhouses in Poland.
Polymerase chain reaction (PCR) assay, restriction fragment length polymorphism (RFLP) analysis, sequencing of 16Sr RNA gene fragments and graft transmission tests were used to investigate the possible association of phytoplasma infection with lily upper leaf necrosis.
PCR-RFLP analysis revealed that lily plants with the same type of symptoms were affected with two distinct phytoplasmas.
The restriction profile of phytoplasma from Catharanthus roseus experimentally infected by grafting using scions of lily Simplon corresponded to that of aster yellows phytoplasma group, subgroup 16SrI-B, now reclassified as Candidatus Phytoplasma asteris. Plants of lily Siberia were infected by phytoplasma classified to apple proliferation phytoplasma group, subgroup 16SrX-A, now reclassified as Candidatus Phytoplasma mali. A comparison of the amplified 16S rDNA sequences of the phytoplasmas from lily Simplon (GenBank accession number EF128036) and lily Siberia (GenBank acession number EF370450) with sequences available in the GenBank confirmed the classification of the phytoplasmas, respectively, as Candidatus Phytoplasma asteris and Candidatus Phytoplasma mali.
