STUDY ON INTERACTION BETWEEN AFLATOXIGENIC AND NON-AFLATOXIGENIC ISOLATES OF ASPERGILLUS FLAVUS

Abstract
Some isolates of A. flavus produce aflatoxin on pistachio seeds but not all of the isolates could produce aflatoxin.
So it seems that study on interaction between aflatoxigenic and non-aflatoxigenic isolates is very important.
In this investigation, three toxic and three non-toxic isolates were determined.
Aflatoxin producing ability was detected on Czapek agar medium with 0.3% β-cyclodextrin.
The production of aflatoxin coincided with the presence of a bright blue fluorescent area surrounding colonies when observed under long wavelength (365 nm) UV light after 3 days of incubation at 26°C. Interaction between isolates was tested by using dual culture method and no antagonistic mechanism was observed among isolates.
Spores of toxic and non-toxic isolates were combined and cultured on test medium.
As a result, non-toxigenic isolates could not reduce aflatoxin value that was produced by aflatoxigenic isolates below the test medium sensitive threshold.
