Abstract
Pollen grain viability and germination are controlled by certain necessary organic and inorganic substances.
In laboratory experiments, commercially produced chemicals are used to resemble the germination conditions in nature.
The objective of this study was to determine the effect of stigma exudate of pomegranate (Punica granatum L. ‘Hicaznar’) on pollen grain germination ratio in vitro.
According to pollen germination tests the agar-in-Petri method, the highest germination of 7.2% was obtained from the medium containing 1% agar + 20% sucrose + 25 ppm boric acid in the beginning of the flowering.
Using the same medium with hanging drop method, pollen grain germination was observed as 19.5% in male flowers while that of 29.1% in hermaphroditic flowers in the beginning of flowering period.
In vitro pollen grain germination tests with hanging drop method were performed with stigma exudate.
Ten stigmas immersed in distilled water by continuous shaking.
The best pollen grain germination was obtained as 18.3% from the stigmas of hermaphroditic flowers immersed in 30 s.
Pollen grain germination was not observed in male flowers in the beginning of the flowering.
Then, one stigma immersed in one drop of water for 30 s was tested.
There was no pollen germination observed neither of the flower type in the beginning of the flowering.
At the end of the flowering period, while the pollen grain germination ratio was 15.1% in male flowers, that of was 3.4% in hermaphroditic flowers.
While the highest pollen viability was observed as 92.4% in male flowers in the beginning of the flowering period, that of was observed as 95.0% in hermaphroditic flowers at the end of flowering period using TTC test.
While a total number of pollen grains per flower were found as 2,143,700 in male flower, that of were found as 1,036,380 in hermaphroditic flowers.
