Abstract
The elimination of Xanthomonas pelargonii and other pathogens succeeds by in vitro-propagation originating from the explanted apex.
The regeneration potency of 31 varieties has been investigated on 26 different media.
On a modified medium acc.
Linsmaier/ Skoog distinct differences between Zonale- and Peltatum- cultivars in the shoot promoting effect of cytokinin compounds were observed.
The highest regeneration rate in Zonale- cv. was induced by a combination of 0.5–1 mg/l IAA and 0.25–0.5 mg/l 2iP; 0.25–1 mg/l BA was less effective.
In Peltatum- cv. the concentration of growth regulators was reduced to 0.1 mg/l IAA and 0.25–0.05 mg/l BA to prevent high callus proliferation.
Lateral and callus born shoots were transferred for root formation to a medium suppl. with 0.5 mg/l IAA. The variety specific regeneration rate varied between 30% and more than 200%. Subcultures of organogenic callus are limited to a few steps because the regeneration potency and the survival rate of adventitious shoots decreased rapidly.
Bacteriological and serological methods for the identification of Xanthomonas in explanted shoot tips and in vitro plantlets were compared.
A scheme for the commercial production of elite stock plants and cuttings with high quality standard is described.
