Microarray analysis of carotenoid and chlorophyll biosynthetic gene expression in the citrus juice sacs irradiated with blue light

Abstract
Citrus fruit is one of the most popular fruit in the world, and represents an important part of our daily diet.
In citrus fruit, high amounts of bioactive compounds are accumulated in the juice sacs.
In the present study, the effects of blue light on the carotenoid and chlorophyll biosynthesis were investigated in the juice sacs in vitro.
During the experimental period, the juice sacs turned orange gradually in the control.
Under the blue light, the changes in the juice sacs colour were more significant than the control during the experimental period.
Interestingly, we found that the juice sacs in the blue light treatment not only turned orange, but also appeared light green colour.
Further analysis of carotenoid and chlorophyll contents showed that the blue light treatment significantly induced the accumulation of β-carotene, β-cryptoxanthin, all-trans-violaxanthin, 9-cis-violaxanthin, lutein, as well as chlorophylls in the juice sacs in vitro.
In addition, to elucidate the roles of blue light in regulating carotenoid and chlorophyll accumulation, microarray analysis of the gene expression was conducted in the present study.
The results showed that the expression of the carotenoid biosynthetic genes (CitPSY, CitPDS, CitZISO, CitCRTISO, CitLCYb1, CitLCYb2, CitHYb, and CitZEP) and chlorophyll biosynthetic genes (CitGGDR, CitHEMA1, CitGSA2, CitHEMB1, CitHEMC, CitHEMD, CitHEME2, CitHEMG1, CitCHLI, CitGHLD, CitCHLH, CitCHLM, CitCHL27, CitDVR, and CitCS) was significantly upregulated by the blue light in the juice sacs during the experimental period.
The high expression levels of the carotenoid and chlorophyll biosynthetic genes in the blue light treatment led to enhance the carotenoid and chlorophyll accumulation in the juice sacs.
The result presented in this study not only contribute to elucidating the roles of blue light in carotenoid and chlorophyll accumulation, but also provide new strategies for improving the quality of citrus fruit.
