Abstract
Electrophoretic results have been used to analyse the genetic constitution and the allel frequencies of populations, to distinguish different cultivars, to recognise hybrids and for measuring relationships between species.
However, the latter application analysing the variation within and between species based on electrophoresis offers serious problems.
The number of recognisable bands is but a part of the total amount of variation encountered in a particular gel.
To solve this problem the following combination of techniques has been used:
However, the latter application analysing the variation within and between species based on electrophoresis offers serious problems.
The number of recognisable bands is but a part of the total amount of variation encountered in a particular gel.
To solve this problem the following combination of techniques has been used:
- an electrophoretic gel was scanned with the aid of a densitometer,
- the heights of the densitograms (one for each individual) were measured on regular intervals,
- the data matrix was transformed and corrected,
- a numerical analysis was performed (i.e. cluster analysis, principal coordinate analysis, principal component analysis).
The features of this method will be discussed.
