Abstract
Ploidy manipulation techniques through protoplast culture was developed for breeding of Japanese persimmon (Diospyros kaki L.). Protoplast isolation conditions were determined for the calli derived from leaf primordia of D. glandulosa and seven cultivars of D. kaki (Jiro, Suruga, Mushirodagosho, Fuyu, Hiratanenashi, Saijo and Miyazakitanenashi). Using agarose-bead culture and modified KM8p medium, dodecaploid plants were obtained by colchicine treatment of ‘Jiro’ (2n=90, hexaploid) protoplasts and protoplast fusion between ‘Jiro’ and ‘Suruga’ (hexaploid).
