DETECTABILITY OF PRUNUS NECROTIC RINGSPOT AND PLUM POX VIRUSES BY RT-PCR, MULTIPLEX RT-PCR, ELISA AND INDEXING ON WOODY INDICATORS

Abstract
The applicability of multiplex RT-PCR for the simultaneous detection of Prunus necrotic ringspot virus (PNRV) and plum pox virus (PPV) was compared to that of conventional RT-PCR assay, ELISA and indexing on woody indicators.
Ten peach, three sour and seven sweet cherry trees were tested for infections.
All samples tested positive in indexing on woody indicators, were also found infected using conventional RT-PCR and multiplex RT-PCR. Using the conventional RT-PCR assays six peach, one sour and three sweet cherry trees were found infected with PPV only.
Two peach, four sweet and two sour cherry trees were found infected with PNRV only.
In two cases, mixed infection with PPV and PNRV was determined.
The multiplex RT-PCR assays gave the same results as the simple, conventional RT-PCR diagnosis, faithfully detecting single or multiple infections by the two viruses.
Our results assure that this RT-PCR assays is applicable for the detection of mixed infections caused by PPV and PNRV as well as showing greater sensitivity than that of ELISA. Multiplex RT-PCR is a promising method for testing a limited number of samples quickly, in order to verify the health status of plant material in certain cases when ELISA results are not reliable.
