THE EUROPEAN PRUNUS MAPPING PROJECT: UPDATE ON MARKER DEVELOPMENT IN ALMOND

Abstract
Four populations are currently being used for map construction with molecular markers in almond.
Two of them are interspecific almond x peach F2‘s: ‘Texas’ x ‘Earlygold’ (‘TxE’), and ‘Garfi’ x ‘Nemared’, and two more are intraspecific almond F1 progenies: ‘Ferragnès’ x ‘Tuono’ (‘FxT’) and ‘D-3–5’ x ‘Bertina’. We have developed detailed maps in two of them: ‘FxT’ with 183 markers (7 isozymes, 120 RFLPs and 56 RAPDs), ‘TxE’ with 106 markers (11 isozymes and 95 RFLPs). The latter population is currently being used to construct a saturated map with 250 markers with the collaboration of other partners in a collaborative European project.
When these maps were compared, a close similarity was observed, not only in the order of the markers, but also in the genetic distance between them.
The other two populations are used for the analysis of specific characters, and we are elaborating maps with a reduced number of markers (3–4 per chromosome). These markers are chosen to cover the whole genome at approximately regular intervals based on the information of the detailed maps.
Analysis of co-segregation between markers and morphological characters has also been performed in these populations, and markers have been localized near genes responsible for shell hardness, self-incompatibility, kernel bitterness, leaf colour and anther colour.
