STUDIES ON DETERMINATION OF ANTAGONISTIC POTENTIAL AND BIOPREPARATION OF SOME BACTERIA AGAINST THE FIREBLIGHT PATHOGEN

Totally one hundred sixty seven epiphytic bacterial strains including Erwinia herbicola, fluorescent pseudomonads, and some Gram(+) bacteria were obtained from the different organs of healthy pear trees in the Aegean Region.
Eleven bacterial strains completely suppressing the development of the disease in immature pear fruit assay (IPFA) were used in pear blossom assay. E. herbicola strains of 78-B/2, 24 and 1B significantly reduced the percentage of blighted blossoms by 82.07% to 98.03% compared to the untreated control.
Streptomycine at 100 ppm did not significantly reduce the blight in the pear blossoms.
Different formulations such as lyophilized, talc-based and whey-based wettable formulations of E. herbicola strains Eh 1B and Eh 24 which were the most promising in in vivo tests against E. amylovora were developed.
The viability of E. herbicola cells in lyophilized formulations were successfully protected for a period of at least 180 days at 24°C and 10°C. In talc-based and whey-based formulations, the bacteria survived even up to 180 days of storage at 10°C although the population began to decline after 120 days.
When the bioformulations were stored at 24°C, the shelf-life of E. herbicola strains in talc-based and whey-based formulations were estimated at 60 days.
Bioformulations were also tested for their ability to suppress the development of E. amylovora on both pear fruits and blossoms.
The bioformulations which were tested on pear fruits suppressed the development of fireblight by 71.43% to 100% compared to the application of pathogen alone.
In the blossom assays different bioformulations of E. herbicola strains reduced the percentage of blighted blossoms by 31.47–84.32 % compared to the application of pathogen alone.
Treatments with talc-based formulations of Eh 1B and Eh 24 were more effective in reducing fruit and blossom blight than the lyophilized and whey-based formulations.
