ANATOMICAL CHANGES IN GLORIOSA ROTHSCHILDIANA O’BRIEN STEMS INDUCED BY JA-ME AND ABA

Abstract
Tuberization of Gloriosa rothschildiana ‘Red Dark’ is inhibited in the presence of different concentrations of ABA and higher levels of JA-Me.
Anatomical changes in stems accompanying the inhibition of tuber development in in vitro culture were studied.
Murashige and Skoog (MS) medium containing 6% of sucrose and 5 mg l-1, 25 mg l-1 JA-Me or ABA at concentration of 0.01 mg l-1 was used.
Stem tissues after a 4-week incubation period on medium were examined.
      Both growth regulators caused the decrease of parenchyma cells size.
JA-Me concentrations invoked the increase of number and diameter of xylem vessels, but ABA a strong reduction of vessel number.
At 25 mg l-1 JA-Me, local necroses in parenchyma were observed.
Histological changes found corresponded with weaker formation of tubers at ABA presence and higher JA-Me concentration.
