Abstract
We attempted to establish biochemical markers for the selection of more winter-hardy genotypes, on the basis of the contents of cryoprotectants (proline and soluble sugars) in in vitro shoot cultures.
The proline and sugar contents were measured in acclimated and non-acclimated shoot explants of six raspberry and two blackberry cultivars.
In experiments on the survival and regeneration after freezing in temperatures from -3 to -18°C, cold acclimated with non-acclimated shoots were compared.
The cryoprotectant contents were higher in the acclimated shoots than in the control.
The rate of increase depended on the initial contents.
The genotypes with a higher level of proline in the non-acclimated shoots produced relatively less proline as a result of hardening, but the content of proline after acclimating was still highest in these genotypes.
A higher percentage of acclimated shoots than non-acclimated shoots survived and differences between genotypes were found.
