Abstract
This study was undertaken with the objective of standardizing the protocol for the micropropagation of strawberry cultivar Oso Grande. Runner tips were obtained from field grown strawberry plants from April to July and were surface sterilized using mercuric chloride (0.1%) for different durations.
A four-minute dip was the optimum duration.
Meristems were cultured on Murashige and Skoog (MS) agar solidified medium supplemented with different concentrations of BA and IBA. Best shoot regeneration response was obtained on MS medium supplemented with 0.5 mg L-1 BA and 0.1 mg L-1 IBA. There was a six- to nearly eight-fold increase in shoot number in six weeks duration.
Proliferated micro-shoots showed 92.3% rooting on half strength medium supplemented with IBA 1.0 mg L-1 after 5-6 weeks of culturing.
The optimum hardening period for strawberry was observed from December to February in polyhouse under Punjab conditions.
Hardening success of 77.3% was observed in rooted plants transferred singly to black polybags (6×10 cm) containing a sterilized mixture of good garden soil and farmyard manure (1:1) after 6 weeks.
