Abstract
Cryopreservation technique is an important tool for germplasm conservation.
Most perennial fruit trees can be successfully cryopreserved, but the regeneration rates are low.
The aim of this study was to evaluate the influence of cold acclimation which consisted of alternating temperatures (25±1°C in daytime/4±1°C at night) or constant temperatures (4±1 and 20±1°C) combined with visual 2, 3, 5-triphenyltetrazolium chloride (TTC) staining test.
The result showed that TTC-staining was efficient to predict the optimal preconditioning treatment.
The study helps us to save plant materials when we optimize the droplet-vitrification protocol.
