TDZ, 2iP and zeatin in blueberry (Vaccinium corymbosum L. ‘Duke’) in vitro proliferation and organogenesis

Abstract
Zeatin and benzyladenine are the most important cytokinins used in commercial plant propagation.
Micropropagation of blueberry cultivars is mostly based on the use of zeatin.
Due to the high costs of zeatin, other growth regulators able to induce high proliferation and regeneration efficiency in different blueberry cultivars, is considered as a priority.
In our study, 2 mg L-1 of zeatin (an example of commercial proliferation media), showed highest axillary bud proliferation of blueberry cultivar ‘Duke’, while elongation of the stems and a reduced proliferation rate was recorded for media without growth regulator.
Thidiazuron (TDZ), an alternative cytokinin-like regulator, alone or in combination with 2iP, induced high shoot proliferation, leaf tissue organogenesis but also high callus formation of the same in vitro blueberry cultivars.
TDZ induced a high callus formation at both concentrations (0.2 and 0.5 mg L-1) tested, but if combined with 2iP (15 mg L-1) callus formation was inhibited and stem elongation promoted.
TDZ can be considered an interesting PGR for improving blueberry in vitro proliferation and regeneration efficiency, especially for the more recalcitrant genotypes.
However, further studies should be carried out in order to verify phenotypic and genotypic stability of the new plants obtained.
